University of Algarve Description Citation | This record contains the analysis code and supporting files for the RNA-seq study " RNA-seq profiling of TRIB2 knockout in human melanoma UACC-62 cell line " by Isabel Duarte, Victor Mayoral-Varo, Rebeca Álvarez, Ana Dopazo, Wolfgang Link, and Bibiana I. Ferreira, submitted to Nature Scientific Data (2026). Background | TRIB2 (Tribbles Pseudokinase 2) is a pseudokinase implicated in the regulation of cell proliferation, survival, and differentiation across several cancer types. This study characterises the global transcriptional consequences of CRISPR/Cas9-mediated TRIB2 knockout in the human melanoma cell line UACC-62 by RNA sequencing, comparing parental and knockout cells across multiple replicates. Raw sequencing data are deposited in ArrayExpress under accession E-MTAB-16439 . The present record contains the downstream analysis and reproducibility materials: run_nf-core_rnaseq_pipeline.sh - shell script used to run the nf-core/rnaseq pipeline for quality control, alignment, and gene-level quantification against the GRCh38 primary assembly reference genome. multiqc_report.html - MultiQC summary report of pipeline quality control metrics. samplesheet.csv - nf-core pipeline input samplesheet specifying sample names, FASTQ file paths, and strandedness. samples.txt - sample metadata file describing experimental conditions and replicate structure. trib2_nat_sci_data.Rmd - R Markdown document containing the code used for the exploratory data analysis and data quality control presented in the Nature Scientific Data publication. trib2_nat_sci_data.html - rendered HTML output of the R Markdown analysis document.
