LONGITUDINAL REJUVENATION STATE OBSERVATORIES AT THE LIMIT English Introduction and Research Abstract Mother DOI: https://doi.org/10.5281/zenodo.22986030 Research Premise This monograph asks a deceptively simple question: when a biomarker becomes younger, what evidence would justify calling the person or system functionally rejuvenated? The answer cannot be supplied by a single biological-age score. A credible claim must survive years of follow-up, predict later function, remain stable across instrument and assay changes, preserve participant and specimen identity, and distinguish durable change from rebound after an intervention stops. The proposed observatory therefore treats rejuvenation as a forecast claim about a partially observed state trajectory. Molecular assays, imaging, wearables, clinical tests, exposure histories, sample handling, and life events are placed on a common chronology without pretending that they share the same time scale or biological meaning. Scientific Boundary The framework does not define any epigenetic, proteomic, metabolomic, imaging, or wearable clock as the true age of a person. It distinguishes responsiveness from validity: a marker may move after an intervention yet fail to predict health, resilience, cognition, mobility, or survival. It also treats drift, sample composition, regression to the mean, informative dropout, and selective follow-up as rival explanations that must be attacked experimentally. Loss of consent, broken identity, inaccessible raw evidence, unresolved safety signals, or irreversible harm are hard vetoes. They cannot be offset by a higher prediction score. Architecture Layer Function 01 Common chronology Align function, molecular assays, imaging, intervention exposure, life events, and sample handling without collapsing their distinct time scales. 02 Independent reference and drift estimation Use replicates, reference materials, cross-instrument checks, frozen bridges, and calibration ancestry to estimate what changed in the measurement system. 03 Reversible and irreversible state models Represent competing trajectories, organ discordance, recovery, rebound, persistent artifact, and irreversible identity cost. 04 Failure injection and adversarial replay Introduce missingness, sample swaps, batch replacements, timestamp shuffles, negative controls, and false rejuvenation signals before trusting performance. 05 Blind future-function adjudication Withhold terminal function, compare against strong clinical baselines, require uncertainty and abstention, and preserve the full decision record for successors. What the Full Research Edition Contains The PDF is organized as a linked research instrument: thirty large gold formula derivations define measurable constructs; eighty-two single-page chapters develop distinct scientific problems; and two hundred single-page Research Gates specify future tests, required evidence, falsifiers, stop rules, and successor handoffs. The gates are designed to be closed by evidence, not by authority. Every chapter opens with a linked formula and points forward to gates. Every gate points backward to the relevant chapter, formula, and DOI evidence anchors. This makes the work readable as a book, auditable as a protocol, and divisible into future studies. Why the Approach Matters Longitudinal intervention studies increasingly report changes in aging biomarkers, but the clocks do not always agree, and movement in a clock does not automatically establish a meaningful health effect. The observatory addresses this gap by requiring held-out future function, strong conventional baselines, negative controls, explicit measurement ancestry, missingness analysis, exposure cessation, and honest abstention. Its governing idea is that the strongest rejuvenation result is not the lowest age estimate. It is a trajectory that another team can replay, challenge, transport to a new setting, and retire if it stops working. Core Candidate Equation O_long = P(F_future | S_t) x I_identity x R_retest - D_drift - B_missing - C_burden The equation is not a universal law. It is a preregistered research design: future-function prediction is multiplied by identity integrity and retest reliability, then penalized for measurement drift, missing-data distortion, and participant burden. Safety, rights, and irreversible harm remain external vetoes rather than terms that can be traded away. Evidence and Citation Policy The research edition strengthens the mother architecture with DOI-resolved primary literature on epigenetic clocks, pace-of-aging measures, longitudinal multi-omics, intervention responsiveness, reliability, physical function, mortality, cell composition, and measurement transport. The mother DOI preserves conceptual lineage; it is not used as a substitute for independent scientific evidence. Selected evidence anchors include: 10.1186/gb-2013-14-10-r115; 10.18632/aging.101684; 10.7554/eLife.54870; 10.7554/eLife.73420; 10.1038/s43587-022-00248-2; 10.1038/s43587-022-00357-y; 10.1038/s41591-026-04562-9; and 10.1038/s41514-025-00312-2. Intended Use This is a research architecture for scientists, observatory designers, reviewers, cohort teams, and successor investigators. It is not medical advice, a diagnostic test, a claim that rejuvenation has been achieved, or authorization to initiate an intervention. Its purpose is to make future claims more testable, more honest about uncertainty, and easier for independent teams to rebuild. The complete PDF should be read as a chronicle rather than a clock: a record of what was measured, when, by which reference chain, under which exposure, with which missingness, and what future function did or did not confirm the claim.
